DNA strands are artificially selected for based on the ability to cleave RNA. The most studied
DNAzyme is the 10 23. The name comes from the 10th round of in vitro selection and the 23rd
round of cloning4. This is a 15 nucleotide sequence with proven enzymatic cleaving properties5. To
target specific sites, RNA recognizing sequences called guide strands that are 7 nucleotides long
are attached to this main catalytic strand on each side. The 10 23 DNAzyme does not require
cofactors, but in some cases they have been shown to improve enzymatic activity6,7.
The challenges facing the development of an antiretroviral drug for Ebola are varied. Effective
cleavage sites need to be found in the genome that are conserved and are attackable based on the
requirement for the 10 23 DNAzyme to cleave. Beyond these targeting barriers, the dosage of the
DNAzyme may depend on the concentration of virions. Furthermore, certain cofactors may
increase the effectiveness of the DNAzyme.
There is has already been some research done in this area to answer these questions. DNAzymes
have already been used exogenously to fight the flu, hepatitis C, and HIV in cultured animal
cells8,9,10. These studies found orders of magnitudes of decrease in viral load upon treatment of
