This lab investigated the central dogma of molecular biology by studying the expression of
jellyfish Green Fluorescent Protein in E. coli (Mardigan, 2011). After transforming E. coli with the
GFP gene under the arabinose operon promoter, GFP expression was analyzed with and without
arabinose. Induction was seen only in media with arabinose. To further study GFP protein and
structure, an SDS Page or gel electrophoresis was conducted where proteins were charged to
allow them to move in an electrical field in terms of their size. To further study the role of the
arabinose operon in GFP expression an overnight culture was done where the plate was frozen and
incubated to induce optimal protein folding. The GFP was then purified. Centrifugation, use of
buffers, and a column was used to separate the GFP and bacterial cells.

The Yupik people wished to reinforce alliances with other groups. At these celebrations the
people related stories, particularly of conflict. The burial ceremony known as the Feast of the
Dead was held by various North American Indian tribes particularly the Iroquois, Huron,
Algonquin, and Ottawa. The ceremony was held on an irregular basis, usually every ten to

Theodore Roethke was born in Saginaw, Michigan on May 25, 1908 to Otto Roethke and Helen
Huebner. Along with uncle, his mother and father owned a local greenhouse, where Roethke
spent a lot of his childhood days working and playing. He referred to the greenhouse as my symbol
for the whole of life, a womb, a heaven on earth ( ). The greenhouse played a huge role in his poetry
. Roethke implies that only after death of the self can people come to realize the true purpose of
life, love. Roethke always had stored in the back of his mind the idea that personal selves were
not the focus of time on earth ( ). Therefore, people must experience the death of their selves
before they can truly live. This idea begins the poem Frau Bauman, Frau Schmidt, and Frau
Schwartze, which begins with the word Gone. Therefore, the poem begins with death although it is
